•Coupled antibody-based enrichment of target analytes with immunodepletion approaches and detected proteins from human plasma previously unseen by mass spectrometry, thereby enabling the possible discovery of new biomarkers
•Tested two different enrichment strategies and their compatibility with mass spectrometry workflows, by spiking proteins into plasma and subsequently enriching them either at protein or peptide level.
•Analyzed the enriched samples via mass spectrometry and the pitfalls of each approach were elucidated.
•Preliminarily, limit of detections as low as ng/mL was uncovered and areas for future optimization were identified to push this limit further.

•Constructed a shuttle vector, pENTR-His-ccdB, that expedites the expression of histidine-tagged recombinant protein and allows testing of different expression systems for uncharacterized proteins.
•Two histidine clones, produced by encoding histidines and tobacco etch virus protease upstream of the recombinant site, was expressed using the newly engineered vector, purified through affinity chromatography and analyzed by western blotting.
Immuno-proteomic analysis of membrane vesicles (MV) from Salmonella enteric serovar Typhimurium (Stm)

•Identified protein components in Salmonella MVs, which are an additional means to induce pathogen-specific immunity, by immunoblotting, two dimensional gel electrophoresis and mass spectrometry
