Research Title: Biochemical Assays for Haemophilus influenzae type B Conjugate Vaccine

Haemophilus Influenzae type b is a small, non motile, gram negative coccobacillus. In the present work I have analyzed the components of the PRP-T conjugate vaccine by several biochemical and analytical assays. PRP-T Conjugate vaccine comprises of PRP (Polyribosyl Ribitol Phosphate) and tetanus toxoid carrier protein. To check the purity of vaccine the amount of free PRP and protein are estimated at different manufacturing stages. The protein content in the bulk, conjugate and filled vials can be estimated by Lowry’s method, modified Lowry’s method and BCA method. The PRP content can be estimated by Orcinol method. The PRP and protein components were found to be within the normal range. LPS (Lipopolysaccharide) is an impurity in conjugate vaccine. I have done modified silver staining technique using SDS-PAGE Electrophoresis to detect LPS. Impurities can also be removed by K Nanocep centrifugal device. Hence by performing different biochemical assays we can check the purity of vaccine.
