“Protective role of &#;-ketoglutarate and withania somnifera (root powder) on hyperammonemic rats : A chronotherapeutic approach”
Short synopsis
Hypothesis: Hyperammonemia is a major contributing factor to neurological abnormalities & oxidative stress mediated tissue damage observed in hepatic encephalopathy and in congenital defects of ammonia detoxication. Currently available drugs/therapies for hyperammonemia are always associated with side effects. Hence, search for a drug with low cost, more active and without side effect is needed. In recent years of scientific investigations, attention has been drawn to the health promoting activity of plants, plant foods and their active components &#;-KG, an intermediate of the citric acid cycle is a potent natural detoxifying agent and Withania somnifera are well known medicinal plants with various traditional medicinal values with the presence of active phytochemicals and multiple biological roles. In the present study, we evaluated the antihyperammonemic effects of &#;-KG and Withania somnifera (WS). Ammonium chloride (NHCl) induced hyperammonemic conditions in Wistar rats. Findings & Conclusions: Administration of &#;-KG and WS restored the levels of ammonia, urea, uric acid, creatinine, non-protein nitrogen, lipid peroxidation products, liver markers, lipid profiles, antioxidants to near normal in hyperammonemic rats. Histological examination of the vital tissues like liver and brain revealed the protective nature of &#;-KG and WS. These findings revealed the antihyperammonemic action of &#;-KG and WS and also &#;-KG and WS effectively scavenge ROS under in vitro conditions showed in vitro antioxidant property of both extracts. Chronotherapeutic effect of W.somnifera at : in hyperammonemic rats may be due to various influencing factors like (i) temporal variations of metabolic enzymes involved in the degradation of phytochemicals and bioflavonoids (ii) temporal variations of lipid peroxidation and antioxidants, lipids, urea cycle enzymes etc., and (iii) temporal variation in bioavailability of W.somnifera. Chronotherapeutic effect of &#;-KG at : in hyperammonemic rats may be due to various influencing factors like (i) the pharmacokinetics of &#;-KG undergoes significant variation in absorption, distribution, metabolism and renal elimination (ii) temporal variations of lipid peroxidation and antioxidants, lipids, urea cycle enzymes etc., and (iii) Timed treatment with &#;-KG acting upon the oscillators in organs, serves to repair the misalignment of oscillators in Hyperammonemia.
